Why Use This Microscope?

This microscope is well suited for viewing samples upright on slides using a SEALED #1.5 coverglass. This instrument is not to be used with dishes or open containers of any sort - these formats should be used on the Leica inverted microscope. All slides MUST be sealed, clean and dry prior to placing on the stage.

Location:

742 Light Hall

Imaging Modes:

Wide-field (conventional) fluorescence and transmitted light, including phase and DIC.

Detector/Cameras:

Princeton Instruments MicroMax CCD camera for low-light, quantitative fluorescence

Photometrics CoolSnap digital camera for color brightfield images, can also be used for multi-color fluorescence, but less sensitive than the MicroMax and not quantitative.

Explanation of size of digital images taken by CCD cameras

Two 35mm film cameras with Zeiss computerized controller

Fluorescence filters: (excitation/emission)

UV/Blue (365 nm for blue dyes such as DAPI and Hoechst)
Blue/Green BP (470 nm for green labels such as Cy-2 and FITC-like dyes) Green/Red (546 nm for Cy-3, Texas Red, or rhodamine-like dyes)

Objective lenses:

    5x / 0.15 Plan-NEOFLUAR
    10x / 0.30 Plan-NEOFLUAR Ph1
    20x / 0.50 Plan-NEOFLUAR Ph2
    40x / 1.30 Plan-NEOFLUAR oil DIC
    63x / 1.40 Plan-APOCHROMAT oil DIC
    100x / 1.40 Plan-APOCHROMAT oil DIC

Charges: $35

Note: To use the microscope you must have an active Account with the Cell Imaging Shared Resource and reserve time using our on-line Scheduling calendar. Contact Carol Ann, Bob, Mary or Sam, to arrange training.
