Research Specialty
Development and application of novel imaging and spectroscopic approaches to studying muscle function.

Research Description
Proper neuromuscular function requires the coordination of multiple body systems, including those for motor command and coordination, energy delivery and utilization, and the generation and expression of force and other mechanical variables. We are using various imaging and spectroscopic approaches to study this integration, in both human and animal studies. These approaches include structural and physiological magnetic resonance imaging (MRI), biochemical studies using magnetic resonance spectroscopy (MRS), tissue oxygenation and blood flow measurements using near-infrared spectroscopy (NIRS), and assessments of motor unit activation using electromyography (EMG). Specific projects include:

Non-invasive Assessment of Muscle Microvascular Function: We have developed an MRI method for assessing changes in blood volume and oxygenation following brief isometric contractions in vivo. We are applying this technique to the study of microvascular dysfunction in persons with Type 2 Diabetes Mellitus.

Biophysical Basis of Muscle Functional MRI: Exercising muscles appear with increased signal intensity in certain types of MR images. These signal intensity changes result indirectly from the metabolic and hemodynamic responses of muscles to exercise. This approach has become known as muscle functional MRI (mfMRI). The objective of this project is to develop a comprehensive and quantitative description of the biophysical and physiological basis of mfMRI, in order to accelerate its application in physiological studies.

Structure-Function Relationships in Skeletal Muscle: The objective of this project is to develop and apply methods for non-invasive study of the relationship between skeletal muscle fibers? architectural properties and mechanical behavior during isometric contraction. In this project, we are combining diffusion-tensor MRI muscle fiber tracking methods, strain imaging, and mfMRI methods.
