Research Description

 

 

Half of clinical acute myeloid leukemia (AML) is preceded by or similar to Myelodysplastic Syndrome (MDS)(also known as preleukemia). This set of diseases is refractory to current treatment except stem cell transplantation (SCT). Because of its age distribution (elderly), most patients are ineligible for SCT. The pathogenesis of these diseases is unknown, but some data suggest a relationship to interstrand crosslink (ICL) DNA damage. The repair system for correcting such damage is largely unknown. We are evaluating genes upregulated in response to ICL damage as genes possiblly relevant to the pathogenesis of MDS and AML.

 

 

Clinical Research Description

 

 

Half of Acute Myeloid Leukemia (AML) cases follow or are biologically related to Myelodysplastic Syndrome (MDS)(also known as preleukemia). The pathogenesis of this set of diseases is unknown, and easily applicable tools to separate these cases from the remainder of AM do not exist. This is important, since treatment difrfers substantially for the two broad groups of AML, MDS-related (MDR-) versus De Novo AML. Interstrand crosslink (ICL) DNA damage is implicated in the pathogenesis of at least some cases of MDS and MDR-AML. Using gene expression, protein microarray, and flow cytometric analysis we are evaluating the cellular response to ICL crosslink DNA damage. Using similar tools we are also evaluating biologic characteristics of bone marrow samples from patients with MDS.
