BONE MARROW ENGRAFTMENT (RFL)

Methodology: Analysis of DNA Polymorphisms for identification of Host and Donor Cells Post Transplantation (Identity/Discrimination Between Samples) by fluorescent PCR and evaluated by capillary electrophoresis
Tube Type: Lavender top (EDTA)
Specimen: Blood or bone marrow, 5-10 mg Chorionic Villi, 2-3 T-25 flasks-cultured cells, paraffin embedded tissue: block, 5-10 unstained slides or 5 curls (NOTE: slides and curls 10m thick with accompanying before and after cut H & E slides); purified DNA (1 g)
Alternate Specimen: Yellow top (ACDH) for whole blood or bone marrow
Pediatric Requirements: 0.5 mL whole blood or bone marrow
Volume: 4 mL (whole blood or bone marrow)
Minimum Volume: 0.5 mL whole blood or bone marrow
Temperature: EDTA and ACDH tubes: room temperature or refrigerated, Chorionic Villi: refrigerate, flasks (cultured cells): room temperature or refrigerate, purified DNA: refrigerate or freeze, paraffin block: room temperature
Stability: EDTA and ACDH tubes - Room temperature: 2 days Refrigerated: 3 days; Purified DNA - indefinitely @ -70C; Paraffin block - indefinitely; Chorionic villi and cultured cells - call Molecular Diagnostics Lab at 615-343-8121
Reasons for Rejection: Decalcification (paraffin embedded tissue). Client will be notified by Molecular Diagnostics Lab if specimen is rejected.
Days Performed: Monday - Friday
Times Performed: Batched
TAT: 3 - 5 days
Significance: Analysis of DNA independently segregating polymorphic sites and 1 gender specific marker for the identification of Recipient versus Donor Cells Post Transplantation
Special Instructions: Call the Molecular Diagnostics Laboratory at 615-343-8121 if additional assistance is needed regarding specimen requirements or submission or if patient has received a transfusion in the prior 2 weeks. Chorionic Villi must be free of maternal cells. Cultured cells - backup culture must be maintained in the event that additional DNA is required.
