The db/db, or Leprdb strain, expresses a spontaneous mutation of the leptin receptor, which results in a point mutation in the cytoplasmic tail of the leptin receptor gene that inhibits receptor signaling. These obese mice represent a model of type II diabetes. db/db mice on the C57BLKS strain are more susceptible to renal disease than the parental C57BL/6. The increased susceptibility may be the result of the more severe hyperglycemia that develops in C57BLKS because of a concomitant insulitis; alternatively, C57BLKS may possess modifier genes that predispose to diabetic nephropathy. Homozygous db/db mice become obese by 3 to 4 weeks of age. Plasma insulin is elevated by 10 to 14 days and blood sugar is elevated by 4 to 8 weeks.
